Plan cloning with the assistant
Plan Gibson, Golden Gate or restriction-ligation cloning in chat, or build a plasmid from parts.
You plan cloning by asking the Design assistant in chat. To build a new plasmid from a backbone and parts, use Design Studio.
Plan an assembly in chat
- Open the plasmid you want to clone into, or @ mention the backbone and inserts.
- Click AI Assistant on the left rail.
- Pick Design on the assistant chip, or press Alt+D.
- Say the method and the parts. For example: "Gibson-assemble the GFP fragment into pET28a."
- Read the predicted product, primers and warnings in the reply.
- To save the result as a plasmid, ask for it. The assistant shows an approval card first. Confirm it to save.
The assistant supports Gibson, Golden Gate and restriction-ligation. For Golden Gate, name the Type IIS enzyme, or BsaI is used. Restriction-ligation needs the enzymes named.
Build a plasmid in Design Studio
- Click New, then New plasmid. Choose Design Studio.
- Work through the four steps: Application, Backbone, Components and Review.
- Choose the exact insertion gap in the backbone.
- Click Prepare draft. A draft opens in its own tab.
- Check the Summary, Changes, Checks and Map tabs.
- Click Confirm and Apply Changes to create the plasmid, or Discard draft.
Click Ask AI on any step to get help from the assistant.
Review open drafts
Drafts also come from the assistant when it swaps one feature for another.
- Open the plasmid. Click Drafts in the plasmid header. It shows the number of open drafts.
- Pick a draft to open its review tab.
- If the source plasmid changed since the draft was made, click Rebase proposal first.
- Click Confirm and Apply Changes or Discard draft.
